Doesn't quite top The 40-Year-Old Virgin, but it comes pretty close. This movie is alternately hilarious and touching. Like Virgin, the problems faced by its main characters are not those brought about by cartoonish villains, but by circumstances and situations. Worth a second viewing for sure.
